942 Inefficient senescent fibroblast clearance in aging skin: The contribution of impaired natural killer cells

چکیده
Cellular senescence, a state of permanent cell cycle arrest, is one the hallmarks tissue aging and age-related disorders. Stromal cells like fibroblasts, present in connective tissues play pivotal role organ homeostasis aging. We previously showed that senescent fibroblasts profoundly accumulated skin during Under some transient conditions, can be successfully removed by innate immune system, such as Natural Killer (NK) cells. here wanted to understand whether were resistant removal NK or alternatively, themselves inefficient remove fibroblasts. Using newly developed mediated killing assay, we line, primary human mouse preferentially killed Primary from old donor (∼70 years) mice (∼700 days) less efficient opposed those young (∼23 (∼100 days), respectively. Of note, there no changes number between individuals. Furthermore, at synapse cells, release granzyme B (GrB) was markedly reduced. Also, content perforin GrB significantly declined. Interestingly, recognition conjugation towards which are crucial for target lysis impaired. In conclusion, these findings suggest reduced ability combined outcome impaired production perforin. This cytotoxicity likely contributes accumulation other organs. These hold promise advance our knowledge on mechanisms underlying may exploited develop novel strategies against

Clearance of senescent decidual cells by uterine natural killer cells in cycling human endometrium
In cycling human endometrium, menstruation is followed by rapid estrogen-dependent growth. Upon ovulation, progesterone and rising cellular cAMP levels activate the transcription factor Forkhead box O1 (FOXO1) in endometrial stromal cells (EnSCs), leading to cell cycle exit and differentiation into decidual cells that control embryo implantation. Here we show that FOXO1 also causes acute senesc...
متن کاملImmunosurveillance of senescent cancer cells by natural killer cells
We recently dissected how senescent tumors can trigger complementing signaling pathways that mobilize natural killer (NK) cells to eliminate malignant cells. In addition to cell-intrinsic effects on proliferation, senescence induces the production of chemokine (C-C motif) ligand 2 (CCL2), which recruits NK cells to mediate direct tumoricidal effects. Hence, senescence activates a cancer cell-ex...
متن کاملIncreased Natural Killer Cells Activity in Schizophrenic Patients
Background: Schizophrenia has been associated with altered immunity. Different studies regarding natural killer cell activity (NKA) in schizophrenic patients have shown inconsistent results. Objectives: To evaluate NK cell activity in schizophrenic patients in comparison with healthy control individuals. Methods: 30 medication-free schizophrenic patients and 41 healthy sex, age and smoking st...
متن کاملThe Effects of Dienogest on Macrophage and Natural Killer Cells in Adenomyosis: A Randomized Controlled Study
Objective Inflammation plays an important role in endometriosis and adenomyosis. Progestin has been used for symptomatic treatment of adenomyosis, although its effect on inflammation has not been studied. We conducted a randomized controlled study to investigate the changes of immune cell expression, i.e. macrophages and natural killer cells, in the tissue obtained from women with adenomyosis w...
متن کاملTHE CYTOTOXICITY PATHWAY OF NATURAL KILLER CELLS IN CORD BLOOD COMPARED TO P ERIPHERAL BLOOD
The cytotoxic activity of natural killer cells is usually tested by radioactive assay CSI Cr release assay), which detects the release of cytoplasmic contents after plasma membrane disintegration of dying cells. In contrast to this indirect evaluation of cytotoxicity, the assessment of cell damage by flow cytometry aims to provide a more exact characterization of the death pathway via detec...
